Prosaposin is a highly conserved 66kD glycoprotein which contains separate domains in tandem for four saposins, A, B, C, and D. These saposins are involved in lysosomal hydrolysis of GM1 ganglioside, gluco- and galactocerebrosides, sulfatides, and sphingomyelin and other sphingolipids. It also exists as a secretory protein in body fluids. Deficiency of PSAP saposin-B, PSAP saposin-C and PSAP saposin-D region are the cause of a variant of metachromatic leukodystrophy, atypical Gaucher disease and Tay-Sachs disease respectively. Alternative splicing results in multiple transcript variants encoding different isoforms. The gene maps to 10q21-q22 region of human chromosome.
Applications
Suitable for use in ELISA, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
ELISA (Sandwich): Detection limit is ~0.1ng/ml as a capture antibody. Immunohistochemistry (FFPE): 5ug/ml Optimal dilutions to be determined by the researcher.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Full length recombinant protein corresponding to aa18-524 of human PSAP (BC001503, AAH01503) with GST tag. MW of the GST tag alone is 26kD.
