Collybistin, also known as ARHGEF9, is an ~60kD cytoplasmic guanine nucleotide exchange factor. It is expressed in the brain and is upregulated at the onset of neuronal differentiation. Collybistin associates with Neuroligin­2, Gephyrin, and GABA receptor subunits to form a complex which is required for GABAergic synaptogenesis as well as hippocampal synaptic plasticity. Mutations in Collybistin are associated with a variety of neurological disorders including epilepsy, anxiety, aggression, mental retardation, and hyperplexia. Human Collybistin contains an SH3 domain (aa8­67), a DH domain (aa103­287), and a Pleckstrin homology domain (aa318­426). Within aa390­516, human Collybistin shares 97% and 57% aa sequence identity with mouse and rat Collybistin, respectively.

Applications
Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilution
Western Blot: 2ug/ml Immunohistochemistry: 8­-25ug/ml in immersion fixed paraffin-embedded sections of human brain (medulla) Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recombinant protein corresponding to Asn390-Lys516 from human Collybistin/ARHGEF9, expressed in E. coli.
