Calmodulin (CaM) is a ubiquitous, calcium-binding protein that can bind to and regulate a multitude of different protein targets, thereby affecting many different cellular functions.

Applications
Suitable for use in Western Blot, Immunoprecipation and RIA. Other applications not tested.
Recommended Dilutions
Western Blot: 0.1-1ug/ml detects calmodulin (~17kD) in RIPA lysates from human A431 carcinoma cells. Previous lots detected calmodulin in RIPA lysates from mouse 3T3 and L6 rat skeletal fibroblasts. Non-stimulated A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with C1035-05. Proteins were visualized using a Gt xMo IgG (HRP) and a chemiluminescence detection system. Immunoprecipitation: 4ug immunoprecipitates calmodulin from A431 RIPA cell lysate. RIA: 10ug/ml detects calmodulin in the range of 200-300ng. Optimal dilutions to be determined by the researcher.
Recommended Secondary Antibodies
I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-14C: IgG, Fc (HRP) Pab Gt xMo I1904-19C: IgG, F(ab’)2 (HRP) Pab Gt xMo
Included Positive Antigen Control
A0001-50: Non-stimulated A431 cell lysate. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide corresponding to the 21 C-terminal amino acids (aa128-148) of bovine calmodulin, conjugated with tyroglobulin
Form
Supplied as a liquid in 0.1M Tris-glycine, pH 7.4, 0.15M sodium chloride, 0.05% sodium azide.
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes both Ca 2+-bound and Ca 2+-free bovine calmodulin. Does not recognize bovine S100-alpha, S100-beta, troponin C or parvalbumin. Species crossreactivity: rat, mouse, human, porcine and wheat germ. Wide crossreactivity expected.
