DOT1L (mammalian counterpart of yeast Dot1) is a 1537aa, 165kD (predicted) nuclear non-SET domain histone H3K79 methyltransferase. It is involved in developmental gene transcription and maintenance of chromatin structure, and may promote mixed lineage leukemia. Within the sequence used as an immunogen, human DOT1L shares 97%, 94% aa identity with mouse and rat DOT1L, respectively. Two human isoforms affect this region: a 1467aa form has an alternate start site at aa71, while a 1513aa form lacks aa89-112. A third isoform of 1739aa has a 202aa C-terminal extension.

Applications
Suitable for use in Immunoprecipitation and Immunohistochemistry. Other applications not tested.
Recommended Dilution
Immunohistochemistry: 8-25ug/ml Immunoprecipitation: 25ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to Gly2-Lys416 from human DOT1L expressed in E. Coli. Species sequence homology: rat (94%) and mouse (97%).

Supplied as a lyophilized powder in PBS, 5% trehalose. Reconstitute with 200ul sterile PBS.
Purity
Purified by Protein A/G affinity chromatography.
Specificity
Recognizes human DOT1L. No cross-reactivity with recombinant human PCDH-8, NME-3, or WISP-3.
