Mast cells contain a number of preformed chemical mediators such as histamine, chymase, carboxypeptidase and proteolytic tryptase. A substantial quantity of tryptase is estimated to be found in mast cells of skin and lung and suggestes this enzyme plays a major role in mast cell mediated events. In vitro studies indicate tryptase can cleave C3 to form C3a anaphylatoxin, inactivate fibrinogen as a coaguable substrate for thrombin and activate latent collagenase. Human mast cell tryptase has considerable potential as a marker for mast cell activation as well as being a mediator of inflammation.

Applications
Suitable for use in ELISA, Western Blot, Immunofluorescence and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
Immunohistochemistry: Frozen sections Immunohistochemistry (Paraffin): 1:10,000. Requires antigen retrieval using heat treatment prior to staining paraffin sections. Sodium citrate buffer pH 6.0 is recommended. Optimal dilutions to be determined by the researcher.

NS1 myeloma cells with spleen cells from Balb/c mice.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Human mast cell tryptase purified from human lung tissue.
Form
Supplied as a liquid in PBS, 0.09% sodium azide.
Purity
Purified by Protein G affinity chromatography from tissue culture supernatant.
Specificity
Recognizes human Mast Cell Tryptase, both alpha and beta isoforms. This antibody is an excellent marker for mast cells and does not bind to any other cell type in Immunohistochemistry. Species Crossreactivity: canine, feline, monkey and rat.
