Occludin is localized at tight junctions of both epithelial and endothelial cells and is highly expressed in kidney and has not been detected in testis. On the cytoplasmic surface of occludin, a domain within the C-terminal 150 residues has been shown to bind directly to the well-characterized tight junction protein zona occludens 1 (ZO-1).

Applications
Suitable for use in ELISA, Western Blot and Immunocytochemistry/ Immunofluorescence. Other applications not tested.
Recommended Dilution
Western Blot: Detects a band at ~65kD. Immunofluorescence (IC): 5ug/ml KO Validated: Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. CACO-2 cells were transfected with Occludin siRNA and reduction of specific signal was observed in immunofluorescence application Optimal dilutions to be determined by the researcher.
Positive Control
T84 cell line (human intestinal epithelium), MDCK cells (canine kidney), Caco-2 cells (human colon adenocarcinoma) and rat liver. Does not appear to detect the most highly phosphorylated form of the occludin protein when tested on Caco-2 cells.
Storage and Stability
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
GST fusion protein consisting of the C-terminal region (~150aa) of human occludin.
Form
Supplied as a liquid in PBS, pH 7.4, 1% BSA, 0.09% sodium azide, 50% glycerol. Labeled with Fluorescein isothiocyanate (FITC).
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes human Occludin Protein. Species Crossreactivity: mouse, rat and canine.
