Pyruvate kinase is an enzyme involved in glycolysis. It catalyzes the transfer of a phosphoryl group from phosphoenolpyruvate to ADP, yielding a pyruvate molecule. There are 4 isozymes of pyruvate kinase in mammals: L, R, M1 and M2. L type is major isozyme in the liver, R is found in red cells, M1 is the main form in muscle, heart and brain, and M2 is found in early fetal tissues as well as in most cancer cells.

Human PKM1/2 is 531aa protein. Human PKM2 389-433aa (IYHLQLFEELRR LAPITSDPTE ATAVGAVEASFKCCSGAIIVLTK) is replaced by (MFHRKLFEELVRASSHSTDLMEAMA MGSVEASYKCLAAAL IVLTE) in PKM1. The PKM2 isoenzyme of pyruvate kinase is specifically expressed at high levels in tumor cells, and can be measured in plasma of patients with advanced breast cancer. The marker is useful for measuring disease activity, sensitivity to chemotherapy and recurrence.
Applications
Suitable for use in Western Blot and ELISA. Other applications not tested.
Recommended Dilution
Western Blot: 1-10ug/ml (chemiluminescence) ELISA: 1:100,000 (50-100ng control peptide/well.) Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O or PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Synthetic peptide corresponding to 16aa of human PK-M1/M2 (Accession # P14618), N-terminal (KLH).
Form
Supplied as a lyophilized powder from PBS, 0.05% sodium azide.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes human PKM1/2.
