MaxLight™ 650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000.

Transport of macromolecules between the nucleus and cytoplasm occurs bi-directionally and is mediated by nuclear pore complexes (NPCs). NPCs are large supermolecular structures that span the nuclear envelope. Although ions, metabolites, and small macromolecules can passively diffuse through the NPC, most larger macromolecules such as proteins and ribonucleoproteins (RNPs) must be transported across the NPC by specific signal-and energy-dependent mechanisms. To date, the import of proteins into the nucleus has been studied in great detail and is considered to be the most well characterized aspect of nucleocytoplasmic transport. Nuclear import is specified by nuclear targeting or nuclear localization sequences (NLSs) in the imported protein.
Applications
Suitable for use in Immunofluorescence, FLISA and Western Blot. Other applications not tested.
Recommended Dilution
Immunofluorescence: 1ug/ml, see reference #8. FLISA: 0.1-1ug/ml Western Blot: 0.1-1ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
Immunogen
A fusion protein consisting of the first 203 amino-terminal residues of the mouse RanGAP-1 protein.

Supplied as a liquid in PBS, pH 7.2. No preservatives added. Labeled with MaxLight™650.
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes both the unmodified (~ 70kD) form and the GMP1 (SUMO-1) conjugated (~90kD) form of the RanGAP-1 protein. The ~70kD form of RanGAP-1 is cytoplasmic, whereas the ~90kD form is associated predominately with the nuclear envelope where it is localized to the nuclear pore complexes (NPCs). Species crossreactivity: human and rat. Lysates Tested: Rat liver nuclear envelopes, total lysates derived from NIH 3T3 and HeLa cells.
