| The Sprouty/Spread family of proteins contains three members, which share high sequence homology for the cysteine-rich domain (CRD), and are shown to inhibit Ras/ERK signaling. Along with its other family members, Spred-2 inhibits Ras/MAP kinase activity (mediated by the epidermal growth factor receptor and the fibroblast growth factor receptor) by binding to Ras and thereby preventing Raf phosphorylation. Like Spred-1, Spred-2 competes with substrates Tau and STAT3 for the DYRK1a kinase domain and thereby inhibits DYRK1a substrate phosphorylation.
