The formation of the cornified (CE) envelope during the late stages of epidermal differentiation is essential for epidermal barrier function and protects the body against environmental attack and water loss. Formation of the cornified envelope involves the replacement of the plasma membrane by cross-linkage of precursor proteins such as loricrin, involucrin and small proline rich proteins (SPRR). SPRR genes constitute a multigene family clustered within the epidermal differentiation complex on chromosome 1q21. This cluster contains two SPRR1 genes, seven SPRR2 genes and a single SPRR3 gene. SPRR gene products are differentially expressed in vivo and in vitro suggesting that the SPRR multigene family evolved to provide highly specialized cornified envelope precursor proteins in stratified epithelia. SPRR2 is mainly expressed in interfollicular epidermis.
