Myogenin is a member of the MyoD family of myogenic basic helix-loop-helix (bHLH) transcription factors that also includes MyoD, Myf-5, and MRF4 (also known as herculinor Myf-6). MyoD family members are expressed exclusively in skeletal muscle and play a key role in activating myogenesis by binding to enhancer sequences of muscle-specific genes. The regulatory domain of MyoD is approximately 70aa in length and includes both a basic DNA binding motif and a bHLH dimerization motif. MyoD family members share about 80% aa homology in their bHLH motifs. Anti-myogenin labels the nuclei of myoblasts in developing muscle tissue, and is expressed in tumor cell nuclei of rhabdomyosarcoma and some leiomyosarcomas. Positive nuclear staining may occur in Wilms' tumor.

Applications
Suitable for use in ELISA, Western Blot, Flow Cytometry, Immunofluorescence, Immunoprecipitation and Immunohistochemistry. Other applications not tested.
Recommended Dilution
Flow Cytometry: 0.5-1ug/10e6 cells Immunofluorescence: 1-2ug/ml Western Blot: 0.5-1ug/ml Immunohistochemistry (Frozen & Formalin fixed paraffin embedded): 0.5-1ug/ml/30min @ RT Immunoprecipitation: 1-2ug/500ug protein lysate Optimal dilutions to be determined by the researcher.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recombinant protein corresponding to human myogenin.
Form
Supplied as a liquid in PBS with 0.05% BSA and 0.05% sodium azide. Also available without BSA and azide, see 138574-AF.
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes human myogenin. Species Crossreactivity: mouse, rat, feline and porcine.
