Thyroglobulin is a 660kD dimeric pre-protein with mutiple glycosylation sites. It is produced by and processed within the thyroid gland to produce the hormone thyroxine and triiodothyronine. Prior to forming dimers, thyroglobulin monomers undergo conformational maturation in the endoplasmic reticulation. The vast majority of follicular carcinomas of the thyroid will give positive immunoreactivity for anti-thyroglobulin even though sometimes only focally. Poorly differentiated carcinomas of the thyroid are frequently anti-thyroglobulin negative. Adenocarcinomas of other-than-thyroid origin do not react with this antibody. This antibody is useful in identification of thyroid carcinoma of the papillary and follicular types. Presence of thyroglobulin in metastatic lesions establishes the thyroid origin of tumor. Anti-thyroglobulin, combined with anti-calcitonin, can identify medullary carcinomas of the thyroid. Furthermore, anti-thyroglobulin, combined with anti-TTF1, can be a reliable marker to differentiate between primary thyroid and lung neoplasms.

Applications
Suitable for use in Flow Cytometry and Immunohistochemistry. Other applications have not been tested.
Recommended Dilutions
Flow Cytometry: 0.5-1ug/10e6 cells Immunohistochemistry (Formalin-fixed): 0.1-0.2ug/ml for 30min at RT. Staining of formalin-fixed tissue requires boiling tissue sections in 10mM citrate buffer, pH 6, for 10-20mins followed by coooling at RT for 20mins. Optimal dilutions to be determined by the researcher.
Positive Control
Thyroid
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Human thyroid follicular cells
Form
Supplied as a liquid in PBS. Also available with BSA and azide, see 138596.
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes a partially defined epitope of human thyroglobulin. Species Crossreactivity: mouse, rat
