Specifications

Human Chemokine (C-C motif) ligand 23 (CCL23) is a small cytokine belonging to the CC chemokine family. CCL23 is also known as myeloid progenitor inhibitory factor MPIF-1, CK8 and SCYA23. CCL23 cDNA encodes a 120aa residue precursor protein with a putative 21aa residue signal peptide that is cleaved to generate a 99aa residue mature CCL23 (aa22-120). Additional N-terminal processing of the 99aa residue variant can generate a 75aa residue peptide (aa46-120) that is significantly more active than the 99aa residue variant. CCL23 binds to CCR1 with high affinity and has chemotactic activity for monocytes, dendritic cells, and osteoclast precursors. CCL23 enhances angiogenesis of endothelial cells, but reduces the proliferation of progenitor cells giving rise to granulocyte and monocyte lineages.

Recombinant Human CCL23 produced in E. coli is a single, non-glycosylated, polypeptide chain containing 100aa with an extra N-terminal Met and having a molecular mass of 11.5kD. The sequence of the first five N-terminal aaa was determined and was found to be Met-Arg-Val-Thr-Lys.

Biological ActivityThe ED50 of rhCCL23 was 0.2-0.5ug/ml as determined by its ability to chemoattract THP1 human acute monocytic leukemia cells. The ED50 of rhCCL23 was 0.02-0.1ug/ml as determined by its ability to chemoattract human CCR1 transfected BaF3 mouse proB cells.
Storage and StabilityLyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile buffer or dH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
