| Mutations in the ABC A1 gene (ATP-binding cassette transporter 1) have been reported in Tangier disease (TD). TD is an autosomal recessive disorder characterized by an absence of plasma HDL, cholesterol ester depositing in the reticulo-endothelial system and deviations in cellular lipid trafficking. ABC A1 mediates the apo-A1 associated export of cholesterol and phospholipids from the cell. It is expressed on the plasma membrane and the Golgi complex, and is regulated by cholesterol flux. Regulation of the cholesterol flux between HDL and macrophages is competitive between ABC A1 and SR-BI. | | | Catalog # | A0004 | | Applications | Suitable for use in ELISA, Western Blot, Immunoprecipitation Immunoflourescence, Immunocytochemistry and Immunohistochemistry. Other applications not tested. | | Recommended Dilution | Western Blot: 1:250. Band representing ABC A1 is seen at ~220kD. Additional non-specific bands are seen at lower molecular weights, but do not interfere with the ABC A1 signal. | | Note: It is important not to boil the sample before loading onto a gel. Boiling can cause aggregation in large proteins, resulting in the protein’s inability to enter the gel. Post-nuclear lysates of 9-cis-retinoic acid and 22R-hydroxycholesterol induced raw macrophages were used for the positive control experiments. | | Immunoprecipitation: See Krimbou and/or Denis, et al. references. | | Immunohistochemistry: 1:100 | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | N/A | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | ~0.5mg/ml | | Form | Supplied as a liquid in Tris-citrate, pH 7.5, 0.02% sodium azide, before the addition of glycerol to 40%. | | Purity | Purified by immunoaffinity chromatography. | | Immunogen | Partial peptide sequence somewhere between amino acids 1100-1300 of the human ABCA1 gene. | | Specificity | Recognizes human ABC A1. Species Crossreactivity: Mouse, rat and hamster. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
