ATF2/7 is a ubiquitously expressed member of the basic region-leucine zipper (bZIP) transcription factor family that regulates gene expression in response to changes in the cellular environment. ATF2/7 plays an important role in placenta formation and skeletal and central nervous system development and is involved in oncogenic transformation and in adaptive responses of the cell to viral infections and toxic stressed. ATF2/7 binds its target promoters/enhancers as a homodimer or as a hetetodimer with a restrictor group of other bZIP proteins. Insulin, epidermal growth factor (EGF) and serum activate ATF2/7 in fibroblasts. The two-step mechanism involves two distinct Ras effector pathways. The first is the Raf-MEK-ERK pathway, which induces phosphorylation of ATF2/7 Thr71. The second pathway inducing ATF2/7 Thr69 phosphorylation requires the Ral-RalGDS-Src-p38 pathway.
