| ADAM10 is a member of the ADAM (a disintegrin and metalloprotease like domain) family. It cleaves the membrane-bound precursor of TNF-alpha at 76-Ala-|-Val-77 to its mature soluble form and is responsible for the proteolytic release of several cell-surface proteins, including heparin binding epidermal growth-like factor, ephrin-A2 and for constitutive and regulated alpha-secretase cleavage of amyloid precursor protein (APP). It contributes to the normal cleavage of the cellular prion protein and is involved in the cleavage of the adhesion molecule L1 at the cell surface and in released membrane vesicules, suggesting a vesicle-based protease activity. | | | Catalog # | A0859-20A | | Cellular Localization | Cell membrane, Type I membrane protein. Also localized in the Golgi apparatus. | | Positive Control | Jurkat whole cell lysate | | Applications | Suitable for use in Western Blot. Other applications not tested. | | Recommended Dilution | Western Blot: 1:500-1:2000. A band at 85kD. A 60kD faint band can be detected insome cell lines including Jurkat, which appears to be the processed mature protein. | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | As reported | | Form | PBS with 0.02% sodium azide | | Purity | Purified by immunoaffinity chromatography. | | Immunogen | Synthetic peptide, corresponding to amino acids 732-748 of Human ADAM10. This sequence is identical to those of bovine and rat origins and differs from that of mouse ADAM10 by one amino acid. Epitope: PQRQRPRESYQMGHMRR | | Specificity | Species Crossreactivity: Crossreacts with Human. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. | | Alternate names | CD156c, HsT18717, kuz, kuzbanian, Kuzbanian Protein Homolog, Mammalian Disintegrin Metalloprotease |
