| ADAM12 is a 120kD type I transmembrane glycoprotein member of the metzincin family of proteases. It is synthesized as a 909aa preproprecursor that contains a 25kD proregion (aa29-207) and a 92kD mature molecule. Proteolytic cleavage in the Golgi removes the propeptide prior to cell-surface expression. A 68kD soluble splice form exists in pregnancy where 36aa replace the C-terminal 205aa (705-909). This circulating ADAM12 isoform remains associated with its prodomain via noncovalent linkage, possibly acting as an enzyme regulator.
