Membrane-bound form of TNF-a is cleaved by a convertase to generate a secreted 17kD mature protein. TNF-a converting enzyme, TACE, is a membrane bound metalloprotease-disintegrin. It also processes other cell surface proteins, including TNF receptor, TGFa, and alpha cleavage of amyloid protein precursor.

Applications
Suitable for use in Immunohistochemistry, ELISA and Western Blot. Other applications not tested.
Recommended Dilution
Peptide ELISA: 1:128,000 Western Blot: 0.1-0.3ug/ml, ~100kD band observed in Human Testis lysates and ~125kD in lysates of cell lines Daudi, HeLa and Jurkat (calculated MW of ~93kD according to NP_003174.3). Primary incubated for 1 hour. Preliminary testing was unsuccessful on Mouse and Rat Immunohistochemistry: 3.75ug/ml on paraffin embedded Human Skin and Thyroid Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Synthetic peptide corresponding to LQRQNRVDSKETEC from the C-terminus of human ADAM17. Species sequence homology: mouse and rat

Supplied as a liquid in Tris saline, pH 7.2, 0.5% BSA, 0.02% sodium azide.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes human ADAM 17. Expected to recognize isoform 1 (represented by NP_003174.3) and not isoform 2 (represented by NP_068604).
