ADAM33 is a widely expressed member of the ADAM family of transmembrane proteins. It contains a metalloprotease-like domain, disintegrin cell adhesion domain, and cysteine rich domain. ADAM33 is involved in the maintenance of airway function, and ADAM33 polymorphisms have been associated with asthma susceptibility and bronchial hyperresponsiveness. Within the mature ectodomain, human and mouse ADAM33 share 79% amino acid sequence identity.
Applications
Suitable for use in Flow Cytometry, Direct ELISA and Immunoprecipitation. Other applications not tested.
Recommended Dilutions
Flow Cytometry: 2.5ug/ml labels 10ul in A549 Human Cell Line. For intracellular staining, cells must first be fixed and permeabilized using 4% paraformaldehyde, 0.1% saponin in PBS. Immunoprecipitation: 25ug/ml to immunoprecipitate recombinant human ADAM33 from conditioned cell culture media. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to Val30-Leu501 (predicted) from human ADAM33, expressed in CHO cells.
Form
Supplied as a lyophilized powder from PBS, 5% trehalose. Reconstitute with 200ul sterile PBS.
Purity
Purified by Protein G affinity chromatography from hybridoma culture supernatant.
Specificity
Recognizes human ADAM33. <5% cross‑reactivity with recombinant mouse ADAM33.
