| Adducin is a heteromeric protein with subunits containing a COOH-terminal myristoylated alkaline-rich C kinase substrate (MARKS-related) domain. The MARKS-related domain, present in a, b, and g adducin subunits, binds calmodulin and contains the major phosphorylation site for protein kinase C (PKC)(3). Adducin is localized at spectrin-actin junctions in erythrocyte membrane skeletons and co-localizes with spectrin at sites of cell-cell contact in epithelial cells(1) and in dentritic spines of hippocampal neurons(2). Studies show that PKC phosphorylation of the MARKS-related domain of adducin regulates actin capping and abolishes the activity of adducin in recruiting spectrin to ends and sides of actin filaments.(3) |

| Recommended Dilution | Immunoblot Analysis: 0.5-2ug/ml detected phospho-Adducin (S724) in lysates from human red blood cells stimulated with PMA. Non-stimulated and PMA-stimulated human red blood cell lysate, was resolved by electrophoresis, transferred to nitrocellulose and probed with A0859-55 (0.5ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. |

| Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
