ADAMTS13 (a disintegrin and metalloproteinase with thrombospondin motifs 13) is a secreted multi-domain zinc protease primarily produced by the liver. In the circulation, it is responsible for cleaving von Willebrand factor (vWF) between Tyr842 and Met843. Defects in ADAMTS13 activity result in the generation of ultra large vWF, leading to thrombotic thrombocytopenic purpura (TTP). The amino acid sequence of human ADAMTS13 in the region of aa34-688 is 76% identical to that of mouse/rat.

Recommended DilutionsFlow Cytometry (Intercellular): 2.5ug labels 10e6 HUVEC human umbilical vein endothelial cells fixed with paraformaldehyde and permeabilized with saponin
Western Blot: 1ug/ml using recombinant human ADAMTS13 under non-reducing conditions only.
Immunoprecipitation: 25ug/ml using conditioned cell culture medium spiked with recombinant human full length ADAMRT13
Optimal dilutions to be determined by the researcher.
Storage and StabilityLyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
