Agrin is an extracellular matrix component which promotes the clustering of nicotinic acetylcholine receptors and other proteins during development at the neuromuscular junction (4). Agrin is a basal lamina protein important in the initial steps of postsynaptic differentiation at the neuromuscular junction. Cloning has revealed that rat agrin is a 220kD protein which contains several different structural motifs. The C-terminal half of agrin also contains 3 sites of alternative splicing referred to as X, Y and Z positions. While no difference in functional activity has been detected between splicing variants at site X (5), insertion of a 4 amino acid peptide at site Y modestly increases agrins nAChR clustering activity. The most dramatic differences in activity between isoforms are observed at splicing site Z. Insertion of an 8aa peptide at this position increases the clustering activity of soluble rat agrin about 10,000 fold (5). Expression of agrin isoforms containing the 8aa or 11aa inserts, or both, is restricted to the CNS (6). In contrast, agrin forms containing no insert at site Z show a widespread expression pattern including muscle. Agrin expression in brain and spinal cord is highest early in development coinciding with the peak of synaptogenesis, and the splicing pattern changes during development (6).
