AIF is a novel mitochondrial intermembrane flavoprotein that is ubiquitously expressed both in normal tissues and in a variety of cancer cell lines. AIF is synthesized as a nonapoptogenic precursor in the cytoplasm and efficiently imported into the mitochondrial intermembrane space. The mature AIF protein is normally confined to the mitochondrial intermembrane space. In a variety of different apoptosis-inducing conditions, AIF translocates through the outer mitochondrial membrane to the cytosol and to the nucleus. Extra-mitochondriaal AIF induces nuclear chromatin condensation and large scale (~50kb) DNA fragmentation. Apoptosis-associated chromatin condensation and degradation is achieved by at least two parallel and redundant pathways. One of these pathways involves caspases, DFF45 (ICAD) and DFF40 (CAD), which leads to oligonucleosomal DNA fragmentation and advanced chromatin condensation. The second pathway is a caspase-independent pathway that involves AIF. These pathways may be activated in a sequential fashion. AIF is a phylogenetically old, bifunctional protein with an electron acceptor/donor (oxidoreductase) function and it has also been suggested to play a role in the regulation of caspase-independent cell death (2,3,4,5).

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
