| Alzheimer’s Disease (AD) is characterized by the presence of extracellular plaques and intracellular neurofibrillary tangles (NFTs) in the brain. The major component of these plaques is Ab peptide (b-amyloid), a 40 to 43 amino acid peptide cleaved from amyloid precursor protein (APP). Increased release of the ‘longer forms’ of Ab peptide, Ab42 or Ab43, which have a greater tendency to aggregate than Ab40, occurs in individuals expressing certain genetic mutations, expressing certain ApoE alleles, or may involve other, still undiscovered, factors. Many researchers theorize that this increased release of Ab42/Ab43 leads to the abnormal deposition of Ab and the associated neurotoxicity in the brains of affected individuals. |

| Catalog # | A2275-73R |
| Applications | Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. |
| Recommended DIlutions | Western Blot: 1:1000 detects 1ug of synthetic amyloid b1-40 polypeptide, but not amyloid b1-42 and 1-43. Amyloid b polypeptides were resolved by electrophoresis, transferred to PVDF and probed with A2275-73R (1:1000). Proteins were visualized using a goat anti-mouse secondary antibody (HRP) and a chemiluminescence detection system. |
| Immunohistochemistry (Paraffin): 1:250 detects amyloid b in human brain sections |
| Optimal dilutions to be determined by the researcher. |
| Recommended Secondary antibodies | I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo |
| I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo |
| Storage and Stability | May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
