Both genetic and biochemical data have established that b-anyloid peptide (Ab) is a causative factor in neuronal death and the consequent diminuation of cognitive abilities observed in Alzheimer’s disease.1,2 Plasma lipoproteins and their cell surface receptors influence sequestration and clearance on soluble Ab, contributing to the etiology of the disease.3,4 Ab may act as an initiating factor in the death of neurons by inducing signal pathways leading to apoptosis.5,6 b-amyloid peptide-binding protein (BBP) is a novel protein containing a structural module related to that of the seven transmembrane domain G protein-coupled receptor superfamily that is important in heterotrimeric G protein activation.7 BBP binds human b-amyloid peptide in vitro with high affinity and specificity. Expression of BBP in cell culture induces caspase-dependent vulnerability to b-amyloid peptide toxicity, suggesting that BBP mediates this cellular vulnerability through a G protein-regulated program of cell death. 7
