Deposition of Amyloid A4 protein precursor (APP) intraneuronally as neurofibrillary tangles, extracellularly as plaques, and in blood vessels is characteristic of both Alzheimerís disease (AD) and aged Downís Syndrome (1). The major protein found in these deposits is a small, insoluble, and highly aggregating polypeptide called amyloid beta peptide (Abeta) that is thought to be derived from aberrant catabolism of its precursor (1). Accumulation of Abeta in the cerebral cortex is a critical event in the pathogenesis of AD (2). APP is cleaved by beta-secretase, producing a soluble derivative of the protein and a membrane anchored 99 amino acid carboxy fragment (C99) (2). The C99 fragment serves as a substrate for an unknown gamma-secretase to generate the 4kD (approximately 4042 amino acids) Abeta which forms the plaque core in AD (3). There are five forms of APP found; APP395, APP563, APP695, APP751, and APP770. They are produced by alternative splicing of the APP gene. APP695 is the most predominant form in neuronal tissue while APP751 is the predominant form found elsewhere.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

ImmunogenHuman APP synthetic peptide corresponding to residues 756-770 of the full length protein or to residues 8599 in the human C99 fragment generated by beta-secretase cleavage. This sequence is identical in mouse and rat over these residues.
SpecificityDetects a ~105kD protein doublet, corresponding to the apparent molecular mass of APP isoforms on SDS-PAGE immunoblots, in samples from human, mouse and rat. An additional band of 35kD, corresponding to the apparent molecular mass of the C99 fragment, is also detected. Species Reactivity: human, mouse and rat
