| The ALK gene is carried by chromosome 2. Upon translocation between chromosome 2 and chromosome 5 [t(2;5}], the ALK gene fuses with the NPM gene. NPM, also known as nucleophosmin or B23, is an ubiquitous nucleolar phosphoprotein involved in the transport of pre-ribosomal components between the nucleolus and the cytoplasm. The chimeric product (NPM-ALK) resulting from t(2;5) translocation is a protein of 80kD with the N-terminal portion of NPM linked to the complete intracellular portion of ALK. ALK is found in many large cell lymphomas. Karpas 299 and SU-DHL-1 lymphoid cell lines strongly express NPM-ALK. | | | Catalog # | A2277-01 | | Applications | Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. | | Recommended Dilution | Immunoprecipitation: 5-10ug/IP reaction | | Western Blot: 1-3ug/ml | | Optimal dilutions to be determined by the researcher. | | Positive Controls | T cell lines with the NPM-ALK translocation, including Karpas 299 and TS cells lysates. | | . | | Storage and Stability: | | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG1 | | Clone No | 3G6 | | Host | Mouse | | Source | Human | | Concentration | ~0.5mg/ml | | Form | Supplied as a liquid in PBS, pH 7.4, 0.1% sodium azide. | | Purity | Purified by Protein A affinity chromatography. | | Immunogen | Recombinant protein derived from the human ALK protein. | | Specificity | Recognizes the human ~80kD ALK protein.
