Ankyrin consists of a single peptide of MW 215kD known to be involved in the attachment of spectrin containing membrane skeleton to the plasma membranes. The cytoplasmic domains of many integral membrane proteins including ion channels and cell adhesion molecules are linked to the actin/spectrin cytoskeletal network by a family of proteins known as ankyrins. Ankyrin belongs to a family of closely related polypeptides associated with the plasma membrane of cells in a variety of cell types (e.g., lymphocytes, platelets, fibroblasts and endothelial cells) and various tissues (e.g., brain, kidney, muscle and many epithelial tissues). Ankyrin molecules contain three distinct domains: an NH2-terminal membrane-binding domain (~89-95kD), a spectrin binding domain (~62kD), and a COOH-terminal domain whose length varies as a result of alternative mRNA splicing. Sequences in the COOH-terminal domain are also the most variable between ankryin family members which include ankyrin R and ankyrin G. Ankryin B or brain-type ankyrin is expressed predominantly as 220kD and 440kD isoforms that are the product of a single gene. Ankyrin G is a marker of axon initial segments, nodes of Ranview, sites of cell-cell contact on epithelial cells and intracellular membranes of some cells.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
