Human AP-2 alpha is a 52kD retinoic acid-inducible, sequence specific DNA binding transcription factor expressed in neural crest lineages. The protein binds to to a GC-rich recognition sequence present in the cis-regulatory regions of several viral and cellular genes, including the enhancers of SV40, HTLV-1, human metallothionein IIa, and mouse major histocompatibility complex. The gene encoding AP-2 alpha is located on chromosome 6 near the HLA locus. AP-2 alpha is required for multiple processes, including limb, eye, craniofacial, cardiovascular, neural tube, and body wall development respectively. It is required for the early morphogenesis of the lens vesicle. It also modulates the function of Myc as a proliferative agent and also as apoptosis inductor. Other known AP-2 alpha-interacting proteins are the retinoblastoma Rb protein, the Yin Yang 1 factor (YY1), and the transcription factors YB-1 and Sp-1. AP-2 alpha may be a factor in predicting survival in patients suffering from epithelial ovarian cancer.
