| The adenomatous polyposis coli tumor suppressor protein homologue APC2 (formerly known as APCL) is involved in the p53/Bcl-2-linked pathway of cell-cycle progression and cell death. APC2 is also heavily involved in the regulation of beta-Catenin transcriptional activity and the formation of beta-Catenin-TCF complexes, and as thus, may also be required for Wnt transduction in all cells. Loss of function mutations in one of the two human APC genes result in improper activation of the Wnt signaling pathway, and are one of the first steps in the development of colon cancer, as well as other tumors. Current research also speculates that APC2 interacts | | | with and may also regulate the cytoskeleton. APC2 bears significant sequence homology to the tumor suppressor APC, and is expressed strongly and specifically in the central nervous system. The expression pattern of APC2 indicates a specific role in proliferation and differentiation in the central nervous system. APC2 is also expressed in several non-neuronal tissues and cell lines including breast, colon and ovary. Overexpressed human APC2 localizes to the perinuclear region and to some microtubles in the cell body.
