| Autophagy is a catabolic process for the autophagosomic- lysosomal degradation of bulk cytoplasmic contents (1,2). Autophagy is generally activated by conditions of nutrient deprivation but has also been associated with a number of physiological processes including development, differentiation, neurodegenerative diseases, infection and cancer (3). The molecular machinery of autophagy was largely discovered in yeast and referred to as autophagy-related (Atg) genes. Formation of the autophagosome involves a ubiquitin-like conjugation system in which Atg12 is covalently bound to Atg5 and targeted to autophagosome vesicles (4-6). This conjugation reaction is mediated by the ubiquitin-E1-like enzyme Atg7 and the E2-like enzyme Atg10 (7,8). | | | Catalog # | A2298-76A | | Applications | Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. | | Recommended Dilution | Western Blot: 1:1000 | | Immunoprecipitation: 1:50 | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | As reported | | Form | Supplied as a liquid in 10mM sodium HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol. | | Purity | Purified by peptide affinity chromatography. | | Immunogen | Synthetic peptide corresponding to residues near the carboxyl-terminus of Atg5. (KLH) | | Specificity | Recognizes endogenous levels of total human Atg5. The observed band represents the Atg12-Atg5 conjugated form, but likely reacts with free Atg5 as well. Species sequence homology: monkey. | | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
