| Beta-arrestin 1 is a member of the arrestin family, which has been implicated in mediating the desensitization and internalization of G protein-coupled receptors (GPCR) (1). Beta-arrestin 1 is constitutively phosphorylated by Erk1/2 at serine 412 of the C-terminus and localized in the cytosol (2). When recruited to the plasma membrane after agonist stimulation of the beta2-adrenergic receptors, beta-arrestin 1 is rapidly dephosphorylated. This dephosphorylation is an essential step of beta-arrestin 1-mediated receptor endocytosis, but it is not required for receptor desensitization (3). | | | Catalog # | A3577 | | Applications | Suitable for use in ELISA, Western Blotting and Immunoprecipitation. Other applications not tested. | | Recommended Dilution | Western Blotting: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight. | | Immunoprecipitation: 1:50 | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG1 | | Clone No | 3A14 | | Host | Mouse | | Source | Human | | Concentration | ~1mg/ml | | Form | Supplied as a liquid in 10mM HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol, 0.02% sodium azide. | | Purity | Purified. | | Immunogen | Synthetic peptide (KLH coupled) corresponding to residues surrounding Ser412 of human beta-arrestin 1. | | Specificity | Detects endogenous beta-arrestin 1 only when phosphorylated at Ser412.
