| Catalog # | A3794-06 | | Description | Mitogen-activated protein (MAP) kinase cascades are activated in response to various extracellular stimuli, including cytokines, growth factors and environmental stresses. A novel MAP kinase kinase kinase (MAPKKK) was recently identified and designated ASK1 (for apoptosis signal-regulating kinase 1) and MAPKKK5 (1-3). ASK1 activated two different subgroups of MAPKK, MKK4 and MKK6, which in turn activated c-Jun N-terminal kinase (JNK) and p38 MAP kinase, respectively. ASK1/MAPKKK5 is activated by TNFR and Fas through the interaction with | | members of the TRAF family and Fas-associated protein Daxx. Overexpression of ASK1 induced apoptotic cell death, and a catalytically inactive form of ASK1 inhibited TNF- -induced apoptosis. ASK1 is expressed in variety of tissues and cell lines. | | Applications | Suitable for use in Western Blot. Other applications not tested. | | Recommended Dilution | Western Blot: 1-2ug’ml | | Optimal dilutions to be determined by the researcher. | | Positive Control | 3T3 cell lysate | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | As reported | | Form | Supplied as a liquid in PBS, pH 7.2, 0.02% sodium azide. | | Purity | Purified by immunoaffinity chromatography. | | Immunogen | Synthetic peptide ASK1 16aa near the amino terminus of | | human ASK1 (Genbank accession No. BAA12684) | | Specificity | Recognizes human ASK1.
