The Na,K-ATPase is an integral membrane protein complex responsible for establishing the electrochemical gradients of Na+ and K+ ions across the plasma membranes of mammalian cells. The enzyme consists of an a and b subunit, both of which exist as isoforms. The a subunit is believed to be the catalytically active subunit, while the b subunit appears necessary for the assembly and transport of the sodium pump to the plasma membrane. The beta subunit is a membrane spanning glycoprotein present in all cell types. The antigenic determinant of this antibody has been mapped to the ectodomain (approximate residues 62-304) of the beta subunit.

ApplicationsSuitable for use in Western Blot and Immunoprecipitation. Other applications have not been tested.
Recommended DilutionWestern Immunoblot Analysis: A 1:2500 dilution detects the b-1 subunit in 50ug of non-reduced brain (~48kD) and kidney (~52kD) microsomal preparations. To reduce background, wash the blot with several changes of buffer containing 0.1% Tween-20, then with a buffer containing 0.5M sodium chloride.
Immunoprecipitation: 5ul immunoprecipitates the b-1 subunit from rat brain microsomal preparation.
Optimal dilutions to be determined by the researcher.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
