Specifications

ATP citrate lyase is the primary enzyme responsible for the synthesis of cytosolic acetyl-CoA in many tissues. The enzyme is a tetramer (relative molecular weight approximately 440,000) of apparently identical subunits. It catalyzes the formation of acetyl-CoA and oxaloacetate from citrate and CoA with a concomitant hydrolysis of ATP to ADP and phosphate. The product, acetyl-CoA, serves several important biosynthetic pathways, including lipogenesis and cholesterogenesis. In nervous tissue, ATP citrate-lyase may be involved in the biosynthesis of acetylcholine.

Assay Condition25ul of ACLY reaction is conducted in a buffer containing 40mM Tris, pH 8.0, 10mM magesium chloride, 2mM DTT, 2mM potassium citrate, 0.1mM CoA, 6-100ng human ACLY at 30ºC for 60 minute. Enzyme activity was assayed using ADP-Glo reagents.
ApplicationsSuitable for use in enzyme kinetics, inhibitor screening and selectivity profiling. Other applications not tested.
Recommended DilutionsOptimal dilutions to be determined by the researcher.
Storage and StabilityAliquot to avoid repeated freezing and thawing. Store at -70°C. Aliquots are stable for 6 months at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
