B7H4, also known as B7x, is a costimulatory protein which is reported to function as a negative regulator of T-cell mediated immunity. Although B7H4 binds an unknown receptor, it is thought to deliver an inhibitory signal to T-cells preventing their proliferation, cell cycle progression and interleukin-2 production. B7H4 deficient mice are only minimally affected; suggesting B7H4 is important in the fine tuning of the T-cell mediated immune response.
B7H4 is expressed on activated T-cells, B-cells, monocytes and dendritic cells. Aberrant expression has been associated with cancers of the lung, breast and ovary in humans. B7H4 could be a useful prognostic marker in Renal Cell Carcinoma (RCC).

ApplicationsSuitable for use in Flow Cytometry and Immunohistochemistry. Other applications not tested.
Recommended DilutionsFlow Cytometry: 1:25-1:50; 10ul labels 10e6 cells in 100ul.
Immunohistochemistry (Paraffin): 1:50. Does not require protein digestion pre-treatment of paraffin sections. Does not require antigen retrieval using heat treatment prior to staining paraffin sections.

Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
