Beta site APP cleaving enzyme (BACE1), also identified as b secretase, is an aspartyl protease. The mature form of the enzyme is an N-glycosylated integral membrane protein with Mr=70kD. The N-glycosylation and carbohydrate trimming of immature BACE1 are observed to take place within the Golgi apparatus. BACE1 cleaves Amyloid Precursor Protein (APP), generating two fragments, the APP N terminal fragment (APPsb) with Mr=100kD and a smaller C-terminal fragment with Mr=12kD (C99). Subsequent cleavage of the smaller fragment by g-secretase produces b-amyloid (Ab). Improperly regulated production of b-amyloid leads to deposition of amyloid plaques found in Alzheimer’s disease, a neurodegenerative, dementia-inducing disorder.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
