| Bad is a 22kD protein that activates apoptosis. It exhibits sequence homology to Bcl-2, a protein that prevents apoptosis, within its BH1 and BH2 domains (1). Bad heterodimerizes with Bcl-Xl and Bcl-2 and reverses the apoptosis suppression effect by Bcl-Xl but not Bcl-2. Bad is phosphorylated by Raf-1, a serine-threonine kinase that is targeted to the mitochondrial membranes by Bcl-2 (2). In the presence of survival factor IL-3, Bad is phosphorylated on two serine residues within its 14-3-3 consensus binding sites. Phosphorylated Bad is then sequestered in the cytosol by binding to 14-3-3 and unable to heterodimerize with Bcl-Xl on mitochondrial membranes to promote apoptosis (3). | | | Catalog # | B0003-46B | | Applications | Suitable for use in Western Blot and Immunohistochemistry.
