BLNK plays a pivotal role in B cell activation through cross-linkage of the B-cell Receptor (BCR), leading to the activation of protein tyrosine kinases (PTKs) including Syk. The resulting tyrosine phosphorylation of BLNK by Syk, provides docking sites for the SH2-containing effector molecules such as Vav, PLC gamma and the adaptor proteins Crb2 and Nck, thereby acting as a central linker protein in many different B cell signalling pathways involved in proliferation, differentiation and apoptosis.

Recommended DilutionWestern Blot: 1:1000-1:2000, detects two bands of approximately 68 and 70kD in mouse Sp2/0 cell lysates.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
