| Bcl-2 is an intracellular non-glycosylated protein that was originally identi ed in the analysis of the (14:18) (q32; q21) translocation break point in follicular lymphoma (1). This translocation results in the juxtaposition of the Bcl-2 gene with the immunoglobulin heavy chain gene locus on chromosome 14 (2). As a result, a high level of Bcl-2 protein is produced (1). The 26kD Bcl-2 protein is localized on the mitochondrial membrane, nuclear envelope, and endoplasmic reticulum (1,3). It regulates cell death (4,5), prolonging cell survival by blocking apoptosis which leads to neoplastic growth without affecting cell proliferation (6). |

| This antibody stains lymphoid tissues (7). In normal lymph node and tonsil tissues, Bcl-2 antibody stains B-cells in mantle zone and interfollicular T-cell areas, but the germinal center cells were not immunoreactive. In the thymus, medullary cells are Bcl-2 positive, whereas cortical cells are negative. In the spleen, Bcl-2 expression was found in both B and T cell areas and in the red pulp, but the germinal center cells were not immunoreactive. Peripheral blood lymphocytes were also strongly positive for Bcl-2 reactivity. Thus, the results indicate that Bcl-2 protein is found mainly in non-proliferating small lymphocytes. |
