| Bcl-6 (also known as LAZ3) is a member of the POZ/zinc finger (POK) family that contains an N-terminal POZ domain and 6 C-terminal zinc finger motifs (1). Bcl-6 is a transcriptional regulator that plays an important role in lymphocyte, myocyte, male germ cells, and keratinocyte survival and or differentiation (2-3). Bcl-6 has also been found to repress a number of genes involved in B cell activation and terminal differentiation, inflammation and cell cycle regulation (4). Chromosomal aberration in Bcl-6 may be a cause of B-cell non-Hodgkin lymphoma and B-cell leukemia (5). | | | Catalog # | B0807-11G | | The gene codes for a 79-kilodalton protein that is homologous with zinc finger-transcription factors. It can interact with a variety of POZ-containing proteins that function as transcription corepressors. It shares amino terminal homology with several transcription factors including the Drosophila tramtrack and Broad complex genes, both of which are developmental transcription regulators. It may be a proto-oncogene specifically involved in the pathogenesis of diffuse large-cell lymphoma (DLCL). It blocks cellular differentiation by repressing transcription of its target genes, and this may promote tumorigenesis. It maps to 3q27 region of human chromosome. It is expressed in germinal center T and B cells and in primary immature dendritic cells. | | Applications | Suitable for use in Immunohistochemistry. Other applications not tested. | | Recommended Dilution | Immunohistochemistry (paraffin):1:10-1:25. High temperature antigen unmasking using 1mM EDTA (pH 8.0) is recommended for formalin fixed paraffin tissues sections. | | Optimal dilutions to be determined by the researcher. | | Positive Control | Tonsil | | Cellular Localization | Nuclear. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG | | Clone No | 8j5 | | Host | Mouse | | Source | Human | | Concentration | Not Determined | | Form | Supplied as a liquid tissue culture supernatant in 0.05% sodium azide. | | Purity | Supernatant | | Immunogen | BALB/C mice were injected with prokaryotic recombinant protein corresponding to amino acids 3-484 of the human bcl-6 protein. | | Specificity | Recognizes human Bcl-6 gene product in follicular lymphomas, diffuse large B cell lymphomas, Burkitt’s lymphomas and in nodular, lymphocyte predominant Hodgkin’s disease.
