| Human Brachyury (transcription factor T) specific PCR primers for RT-PCR (reverse transcription | | | followed by polymerase chain reaction) analysis of mRNA expression. | | | Catalog # | B2674-75B | | The Brachyury protein is required for the proper formation and differentiation of posterior mesoderm and for axial development in all vertebrates. The Brachyury gene encodes a transcription factor that binds to a specific DNA element via its N-terminal region. A protein motif within the DNA-binding domain, the so-called T box, is highly conserved among T homologs from different species and also defines a broader family of T-box genes Brachyury functions during the earlier stages of gastrulation, which suggests that at least one of its early roles is to regulate the morphogenetic behavior of nascent mesoderm by altering cell autonomous properties. | | Components | Primer Pair: Lyophilized. Each vial contains 375pmoles of each primer. Adjust to a final concentration of 7.5 pmoles/ul by resuspending Primer Pair in either 50ul autoclaved deionized water or 0.1X TE buffer (1mM Tris HCl, pH8.0 @ 25°C; 0.1mM EDTA, pH 8.0 @ 25°C). | | Positive Control 56 | Lyophilized. Each vial contains 150 ng synthetic double-stranded DNA. Resuspend in 30ul autoclaved deionized water or TE buffer. To prevent contaminating samples and reagents, it is strongly recommended that the Positive Control is resuspended in a separate location from where PCR reactions are set up. Use a different pipette than one used for PCR set up. The Positive Control is not intended for quantitative purposes. | | GenBank Accession Number | NM_003181 | | Product Sizes | cDNA: 430 base pairs (bp) | | Genomic DNA: 3387 bp product predicted. No product observed. | | Positive Control 56: 320 bp | | Alternate splice variants: None reported. | | Suggested Reaction Conditions | For each 50ul Polymerase Chain Reaction mixture, 2ul of the primer pair stock solution should be used. Sufficient reagent is supplied for 25 separate amplification reactions of the gene fragment. | | Thermocycling Conditions | A typical 50ul PCR would involve a 4 minute denaturation step at 94ºC, followed by 30-35 cycles of 45 seconds at 94ºC, 45 seconds at 55ºC and 45 seconds at 72ºC, with a final 10 minute extension step at 72ºC. | | Storage and Stability | The Primer Pair resuspended in water or TE buffer is stable for up to one year at -20°C in a non-frost free freezer. Aliquot in single use portions. Do not use past the expiration date above. Avoid repeated freeze-thaw cycles. | | Important Note | This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
