| Brain-derived neurotrophic factor (BDNF) is a member of the neurotrophin family of growth factors that includes NGF, NT-3, and NT-4. All neurotrophins have six conserved cysteine residues and share a 55% sequence identity at the amino acid level. BDNF has been shown to enhance the survival and differentiation of several classes of neurons in vitro, including neural crest and placode-derived sensory neurons, dopaminergic neurons in the substantia nigra, basal forebrain cholinergic neurons, hippocampal neurons, and retinal ganglial cells.1 BDNF is expressed within peripheral ganglia and is not restricted to neuronal target fields, raising the possibility that BDNF has paracrine or even autocrine actions on neurons as well as non-neuronal cells.2 | | | Catalog # | B2700-02L | | Cellular Localization | Secreted | | | | Applications: | | Suitable for use in ELISA and Western Blot. Other applications not tested. | | Recommended Dilution | ELISA: Detects 12.5ng/well of rhBDNF. | | Western Blot: detects BDNF at 300ng/lane under non-reducing and reducing conditions. | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | | | May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Monoclonal | | Isotype | IgG1 | | Clone No | 8J359 | | Host | Mouse | | Source | Human | | Concentration | ~0.5mg/ml | | Form | Supplied as a liquid in PBS, pH 7.2. | | Purity | Purified by Protein A affinity chromatography. | | Immunogen | Recombinant human BDNF (rhBDNF) expressed in Sf 21 insect cells. | | Specificity | Monoclonal Anti-Human BDNF will neutralize the biological activity of rhBDNF.
