MUC16/CA125 antigen is a serum marker that is used routinely in gynecologic practice to monitor patients with ovarian cancer. It is a mullerian duct differentiation antigen that is overexpressed in epithelial ovarian cancer cells and secreted into the blood, although its expression is not entirely confined to ovarian cancer. Most biochemical studies have concluded that MUC16/CA125 is a high molecular mass glycoprotein.

According to the ISOBM-TD-1 workshop, CA125 contains three antigenic domains A-domain (Oc125), B-domain (M11), C-Domain (Ov197). The CA-125 antigen is present on a mucin-like glycoprotein of >200kD and is detectable on the apical surface of normal endometrium and endocervix.
ApplicationsImmunohistochemistry: (frozen) 1:10-1:40, (paraffin) 1:10-1:40.
Requires antigen retrieval using heat treatment prior to staining of paraffin sections.
Optimal working dilutions to be determined by researcher.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

ImmunogenOVCA433 ovarian cancer cell line.
SpecificityReacts with an antigen common to most non-mucinous epithelial ovarian carcinomas. Staining is positive in serous and endometrioid adenocarcinomas, Krukenberg's tumor and uterine myoma. Mucinous tumors and normal ovarian epithelium are negative.
