CA125 determinant is present on a mucin-like glycoprotein of high molecular weight. CA125 has been found on frozen sections of amnion and derivatives of coelomic and mullerian epithelium, including pleura, pericardium and peritoneum. In adult tissues, epithelial cells of fallopian tube, endometrium and endocervix, pancreas, colon, gall bladder, stomach, kidney, apocrine sweat gland and mammary gland.

MUC16/CA125 antigen is a serum marker that is used routinely in gynecologic practice to monitor patients with ovarian cancer. It is a mullerian duct differentiation antigen that is overexpressed in epithelial ovarian cancer cells and secreted into the blood, although its expression is not entirely confined to ovarian cancer. Most biochemical studies have concluded that MUC16/CA125 is a high molecular mass glycoprotein.
According to the ISOBM-TD-1 workshop, CA125 contains three antigenic domains A-domain (Oc125), B-domain (M11), C-Domain (Ov197). The CA-125 antigen is present on a mucin-like glycoprotein of >200kD and is detectable on the apical surface of normal endometrium and endocervix.
ApplicationsSuitable for use in Immunohistochemistry. Other applications not tested.
Recommended DilutionImmunohistochemistry (Formalin/paraffin): 1:200 for 30 minutes. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 minutes followed by cooling at RT for 20 minutes.
Optimal dilution determined by the researcher.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
