| Cadherins are a family of CA2+ dependent intercellular adhesion molecules that play a central role in controlling morphogenetic movements during development. Their function is regulated by association with the actin cytoskeleton by a complex of cytoplasmic proteins called the catenin (alpha, beta and gamma). Members of the cadherin family include Cadherin 6 (K-Cadherin), P-cadherin (MW 120kD), E-cadherin (uvomorulin), N-cadherin (neural cadherin), R-Cadherin, cadherin 5, L-CAM and EP-Cadherin. | | | P-cadherin is an 882 amino acid polypeptide with a putative signal peptide, a putative precursor region, an extracellular domain containing internal repeats, and a highly hydrophobic transmembrane region. Amino acid sequences among P-, E- and N-cadherin are most conserved in the cytoplasmic domain followed by conservation of the N-terminal region of the extracellular domain. The cytoplasmic domain provides a link to the cytoskeleton through the associated catenin proteins. The mouse P-cadherin genes have been shown to be tightly linked on chromosome 8. The human E-cadherin gene has been localized to chromosome 16p11-16qter. Changes in P-cadherin expression have been implicated in breast and melanocytic tumors. | | | Catalog # | C0108-37A | | Applications | Suitable for use in Immunohistochemistry. Other applications not tested. | | Recommended Dilution | Immunohistochemistry (Formalin/paraffin): 1:20 for 30 min at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min. | | Optimal dilutions to be determined by the researcher. | | Positive Control | Skin | | Storage and Stability | May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months.
