Calsequestrin is a major Ca2+ binding protein of the sarcoplasmic reticulum (SR) that is found in the terminal cisternae of the SR. It is an acidic protein with a large capacity (4050 moles of Ca2+ per mole of protein) but moderate to low affinity (1mM) for Ca2+ (1,2). Calsequestrin apparently buffers free Ca2+ in the cisternae of the SR and thereby reduces the gradient against which the Ca2+-ATPase must pump Ca2+. It also localizes Ca2+ at the junctional face of terminal cisternae for rapid release into the cytoplasm via the ryanodine receptor (3). Mammalian isoforms of calsequestrin specific for cardiac and skeletal muscle have been identified (4,5).

ApplicationsSuitable for use in Western Blot, Immunoprecipitation, Immunocytochemistry and Immunofluorescence. Other applications not tested.
Recommended DilutionWestern Blot: 0.5ug/ml detected the 60kD calsequestrin in sarcoplasmic reticulum preparation from rat heart ventricles.
Immunoprecipitation: 5ug/ml immunoprecipitated calsequestrin from a lysate of sarcoplasmic reticulum preparation from rat heart ventricles.
Immunocytochemistry: 10ug/ml detected calsequestrin in frozen sections of rat heart by immunofluorescence.
Optimal dilutions to be determined by the researcher.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C.
