| Calsequestrin is a major Ca2+ binding protein of the sarcoplasmic reticulum (SR) that is found in the terminal cisternae of the SR. It is an acidic protein with a large capacity (40–50 moles of Ca2+ per mole of protein) but moderate to low affinity (1mM) for Ca2+ (1,2). Calsequestrin apparently buffers free Ca2+ in the cisternae of the SR and thereby reduces the gradient against which the Ca2+-ATPase must pump Ca2+. It also localizes Ca2+ at the junctional face of terminal cisternae for rapid release into the cytoplasm via the ryanodine receptor (3). Mammalian isoforms of calsequestrin specific for cardiac and skeletal muscle have been identified (4,5). |
