CaMKII, one of the most abundant neuronal protein kinases was first identified in neuronal membrane fractions. CaMKII phosphorylates a broad range of substrates and is expected to have pleiotropic regulatory actions in cellular differentiation and mature neuronal functions. This enzyme is composed of four different chains: alpha, beta, gamma, and delta. The alpha subunit of CaMKII is identified as the major postsynaptic density protein (mPSDp). It undergoes calcium/calmodulin-dependent autophosphorylation on Thr286 in its regulatory domain, rendering the kinase partially calcium-independent. Thus this Thr286 autophosphorylation controls subcellular targeting of CaMKII in neurons with important functional consequences. The protein is enriched in brain and its overexpression alters the general cell growth, differentiation, and morphology of neurons. human CaMKII gene maps on chromosome 5 at 5q33.1.
