| CaMKII, one of the most abundant neuronal protein kinases was first identified in neuronal membrane fractions. CaMKII phosphorylates a broad range of substrates and is expected to have pleiotropic regulatory actions in cellular differentiation and mature neuronal functions. This enzyme is composed of four different chains: alpha, beta, gamma, and delta. The alpha subunit of CaMKII is identified as the major postsynaptic density protein (mPSDp). It undergoes calcium/calmodulin-dependent autophosphorylation on Thr286 in its regulatory domain, rendering the kinase partially calcium-independent. Thus this Thr286 autophosphorylation controls subcellular targeting of CaMKII in neurons with important functional consequences.
