The carcinoembryonic antigen (CEA) is a member of a large family of glycoproteins and a useful tumor marker for adenocarcinoma. In the past, clinical utility of CEA antibodies in immunopathology has been limited by crossreactivity with CEA-related antigens, particularly Non-Speci c Cross-Reacting Antigen (NCA), in normal colon mucosa and human granulocytes.

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

SpecificityReacts speci cally with a restricted epitope on the carcinoembryonic antigen (CD66e, 180kD) in the gastrointestinal tract and does not react with normal colon tissue. Unlike most other CEA antibodies, does not react with Non-Speci c Cross-Reacting Antigen (NCA or CD66c, 55kD.) Col-1 is an established marker for gastric and colon adenocarcinoma. In a study of 35 different types of normal tissues, zero demonstrated positive immunoreactivity. Of 59 tested benign and malignant tumor tissues, reactivity was observed only in neoplasms of epithelial origin, particularly those from the gastrointestinal tract, breast, lung and bladder. Certain pre-malignant cells, such as colonic villous adenoma, also expressed this antibody.
