| B-catenin is a key downstream effector in the Wnt signaling pathway (1). It is implicated in two major biological processes in vertebrates: early embryonic development (2) and tumorigenesis (3). CK1 phosphorylates B-catenin on Ser45. This phosphorylation event primes B-catenin for subsequent phosphorylation by GSK-3 (4-6). GSK-3B destabilizes B-catenin by phosphorylating it at Ser33, Ser37 and Thr41 (7). Mutations in these phosphorylation sites, which result in the stabilization of B-catenin protein levels, have been found in many tumor cell lines (8). | | | Catalog # | C2069-50Q | | Applications | Suitable for use in Western Blot. Other applications not tested. | | Recommended Dilution | Western Blot: 1:1000. Incubate membrane with diluted antibody in 5% w/v BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight. | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | As reported | | Form | Supplied in 10 mM sodium HEPES (pH 7.5), 150mM NaCl, 100ug/ml BSA, 50% glycerol and less than 0.02% sodium azide. | | Purity | Purified by Protein A affinity chromatography. | | Immunogen | Synthetic peptide corresponding to a region surrounding residue Ser37 of human B-catenin (blue carrier-coupled). | | Specificity | Recognizes human non-phospho-B-Catenin (Ser33/37/Thr41).
